Printers differ: some print on the side facing up in the tray, others on the side facing down. Mark an X on a plain sheet, print once and look before you load labels.
Hold a plain-paper print over a 22805 sheet against a window. If it sits off, calibrate once and save the correction for that printer.

Yes. Set "Start at label" to the first unused label: label 1 is the top left, and the count runs across each row. The labels before it stay blank.
Check that the print dialog says 100% (Actual size), not "Fit to page". If the print still sits a little off, print the test sheet on the calibration page and save a correction for your printer.
